Thank you for raising this issue.
As you found out this feature is not currently supported in Netplan and
I'm unclear if it's supported in NetworkManager/nm-applet, yet. I think
it's a reasonable feature request and tagged it as an upstream Wishlist
item.

Bug description:
I am currently trying to debug why I can use a nameserver with a
nondefault (i.e. != 53) port for wireguard connections with wg-quick
and systemd.netdev, but not with Network-Manager and the nm applet
with Lubuntu and Xubuntu. The editor does not accept the syntax.
But:
base on the manpage of netplan (see "nameservers"), it does not accept
a port number, just an ipv4 or ipv6 address.
So not all network configurations can be used with netplan / nm-
applet.
