the same thing happened to me.So I uninstalled the mplayer from synaptic
and installed it from svn sources an it worked.
the output of 
'ldd `which mplayer` | grep directfb`
shows me this
libdirectfb-1.2.so.0 => /usr/lib/libdirectfb-1.2.so.0 (0x00781000).
I tried to help you as I saw you have karmic i386 installed.
I updated my jaunty to karmic and the installation from svn worked.
to install it from svn:
#mkdir mplayer
#cd mplayer
#svn checkout svn://svn.mplayerhq.hu/mplayer/trunk mplayer
#svn update
#./configure --enable-gui
#make
#sudo make install
